Where is best to start?

You could fit a chain tensioner in place of the rear derailleur.
If you have a rear hub which takes a cassette you could buy a single speed conversion kit containing a sprocket and some spacers. Of course you would need to remove your shifters and possibly buy some shorter chainring bolts

If you have the wheel already then buy a cassette or freewheel conversion depending on type. Saves buying a new wheel :)

I mentioned running a tensioner as I thought this was the mountain section and was a mountain bike you were doing! Doh :facepalm: .

I agree with Nick you should be able to run the setup without a tensioner if you have horizontal dropouts.
