As Perry said, there are so many variables.

Rather than going backwards and forwards trying different size sprockets
etc., get out on your bike and select a gear that you are comfortable with, then calculate what size gear it is ( in inches ) using one of the on-line gear calculators.
All you do then is select your singlespeed chainring/sprocket combo to suit the same size gear.
 
For off-road use either 32T or 34T chainrings. I prefer 32T if for no other reason than they give a bit more log/rock clearance.

32:16 is the "standard" off -road ratio ( and a good starting point maybe)but a lot depends on where you ride, whether you have much flattish terrain or road work to do, how steep or technical your trails are.

I run 32:18 most of the time - if you can spin a high cadence it's ok for 14mph or so on the road or flat trails yet it's deep enough for some tough climbs.
I would suggest buying a decent deep tooth 32T chainring and two or three cheap (like the Gusset ones) SS rear cogs in maybe 16, 18, 20T and just play about with the gearing. You can always get better ones once you decide what suits you the best.
 
Shameless plug, look at my for sale topic, single speed kit with 14, 16, 18t and 20t rings for £10. That coupled with a few front ring sizes will surely get a good ratio.
 
Try using your middle or big chain ring and decide which cog on the rear cassette you can get away with staying in. Just about nailed it for me and my commuting on flat but offroading would be much more variable, currently got 40-16 on my BSX play bike, yet to try off road
